Guggul essential oil (GEO) derived from Boswellia serrata is utilized in Ayurveda as traditional and complementary medicine. However, because of their chemical instability, volatile in nature, strong aromas, and poor absorption, the use of GEO is restricted. The objectives of this work were to create Guggul essential oil-based O/W (oil/water) nanoemulsions (GNE) and evaluate their biological potential. GNE were prepared by using Tween-80, Ethanol and SDS using ultrasonicator. Through the use of Particle Size Analyzer, UV, fluorescence, and FT-IR methods, physiological characterization was completed. Numerous biological activities, such as anti-inflammatory, anti-diabetic, anti-bacterial and anti-cancer potential was estimated. The spherical-shaped nanoemulsions had globule sizes of 220 nm, a zeta potential of -35.30mV, a polydispersity index of 0.25, and a drug entrapment efficiency of 96.5% (w/v). GNE's in vitro drug release occurred slowly roughly 45% at six hours. GNE exhibited remarkable antioxidant potential such as: 46% for scavenging DPPH radicals, 33% for scavenging ABTS radicals, 20% for scavenging hydroxyl radicals, and 81% for the iron chelating test. Antibacterial activity was observed with inhibition zone ranged from 0.3 to 1.1 cm against G+ and G- bacteria. After being exposed to 5 µl of GNE, 20% of HeLa cells were viable. GNE exhibited 70% inhibition of α-amylase activity at its higher dose of 250 µL. GNE remained stable even for 50 days. Kinetics of drugs showed that nanoemulsions displayed the Higushi model. GNE potential in the pharmaceutical, food industry has also been explored.
